Selank was developed at the Institute of Molecular Genetics (Russian Academy of Sciences) in cooperation with the V.V. Zakusov Research Institute of Pharmacology (Russian Academy of Medical Sciences), along with another nootropic drug Semax. Selank has been approved in Russia for the treatment of generalized anxiety disorder (GAD) and for its use as a nootropic.

Selank is a synthetic analogue of the immunomodulatory peptide Tuftsin. Both Tuftsin and Selank have the ability to reduce anxiety and increase serotonin levels.

Selank belongs to the group of anxiolytics and nootropics, significantly suppresses anxiety, and depression, has antidepressant effects, and may be useful in the treatment of generalized anxiety disorder (GAD), but also has shown an improvement in cognitive function, learning ability, and memory. Selank has an advantage over traditional anxiety treatments, such as benzodiazepines, as it has no sedating or negative cognitive side effects and no associated addiction or withdrawal problems.

Selank mimics many of the effects of tuftsin that can be used in pharmacology to suppress the side effects of stress and anxiety, improve cognitive functions and brain functions, or strengthen the immune system. For example, Selank induces the metabolism of serotonin, influences the concentration of monoamine neurotransmitters or modulates the expression of Interleukin-6 (IL-6), and affects the balance of T helper cell cytokines. While most prescription drugs cause many undesirable side effects, previous clinical trials and Selank research have concluded that this peptide has no negative side effects or risk of addiction.

Serotonin or 5-hydroxytryptamine (5-HT) is a monoamine neurotransmitter. It has a popular image as a contributor to feelings of well-being and happiness, though its actual biological function is complex and multifaceted, modulating cognition, reward, learning, memory, and numerous physiological processes such as vomiting and vasoconstriction.

Research in patients with depression indicates that Selank can suppress the gene responsible for the production of the inflammatory cytokine IL-6. Interestingly, this effect is only seen in patients with depression and does not appear to take place in healthy individuals. This suggests that Selank may be useful in treating people with anxiety-asthenic disorders, severe disorders in which anxiety is associated with fatigue, headache, heart palpitations, high blood pressure, nerve pain, and depression.

When comparing Selank to standard anxiolytic treatments, like benzodiazepines, the two treatments show similar benefits in reducing anxiety, but only Selank has any effect on asthenic symptoms like fatigue and pain.

Part of the effect is likely due to Selank’s ability to modulated IL-6 expression while part is likely due to the peptide’s ability to alter the rate of breakdown of the body’s natural pain killers, enkephalins.

There has long been an association between anxiety and learning/memory. The correlation is a negative one, with increased anxiety leading to decreased ability to recall memories or store new information. Traditional anxiety treatments can reduce this effect, as can Selank, but Selank appears to do more than simply reduce the impact of anxiety on cognitive function. There is good reason to believe that Selank actually boosts cognition directly.

Research even suggests that Selank can help to rescue memory and learning following damage to the brain. Rats treated with a neurotoxin and then Selank showed restored cognitive processes in at least one study. This effect appears to be related to an artificial inhibition of the catecholamine system in the brain by Selank.

There is some hope that Selank will help to shed light on how cognitive function can be restored or at least improved following traumatic brain injury, particularly the kinds of injury that occurring during the birth process.

In clinical trials, Selank rapidly increased brain neurotrophic factor (BDNF) expression in the hippocampus of rats: BDNF acts on certain neurons of the central nervous system and the peripheral nervous system, helping to support the survival of existing neurons, and encouraging growth and differentiation of new neurons and synapses. BDNF itself is important for long-term memory.

Scientific studies and clinical trials to date have not shown any significant side, undesirable or toxic effects when investigating the Selank peptide.
